Syllabus of Special Methods of Instrumental Analysis (practical classes)
Potentiometry with ion selective electrodes - determination of low concentration of iodides (impurity) in sample of sodium chloride, measurement selectivity, method of two standard additions and its numeric evaluation.
Stripping voltammetry – determination of traces of impurities (heavy metals) – identification and determination in sample of sodium chloride using standard addition method.
Capillary zone electrophoresis - determination of sorbic and benzoic acids in unknown sample, calibration.
Capillary isotachophoresis – determination of glucosamin in food supplement, comparison with standard solution with known active substance concentration, contactless conductivity detection.
UV spectrophotometry – selective determination of constituents of multi-component mixtures (o-hydroxybenzoic and p-hydroxybenzoic acids), selection of optimal wavelengths, evaluation of absorption coefficient, determination of components in unknown sample.
High-performance liquid chromatography – identification of components in mixture of active substances (paracetamol, p-hydroxybenzoic acid, acetylsalicylic acid + product of decomposition – salicylic acid), determination of paracetamol, identification of impurity, decomposition of acetylsalicylic acid.
Sequential injection analysis – automation of nitrates determination using derivatization reaction in the system of sequential injection analysis, optimization of flow rate, „stop-flow“ technique for spectrophotometric determination, calibration, determination of nitrates in drinking or surface water.
Fluorimetry – derivatization of fluorimetric determination of thiamine after oxidation in alkaline conditions, selection of excitation and emission wavelengths, calibration, determination of thiamine in unknown sample.
Raman spectrometry – identification of methanol in samples of ethanolic solutions, determination using comparison of band area of methanol and ethanol against low signal in different part of Raman spectrum.
The aim of the subject is based on practical classes of selected instrumental techniques from the perspectives of practical applications, advantages and also limitations of the use for analysis of different samples. Practical classes are organized in groups of 4 - 5 students.
