Information on teaching this subject will be sent to students electronically (usually at the beginning of the 3rd week of the semester) after closing the SIS for student enrollments when the total number of students enrolled for this subject is already known.The practice of advanced spectrometric methods currently consists of three compulsory tasks (solved in any order), which are carried out in the laboratories of the Department of Analytical Chemistry at Charles University and in the Laboratories of the Department of Trace Element Analysis, Czech Academy of Sciences. Individual tasks will be realized in terms and times given by each supervisor. List of tasks Kinetic assays with spectrophotometric detection. (a task for 2.5 hours) Determination of total mercury content (AMA-254) and speciation analysis (HPLC-ICP-MS) in biological matrix samples. (a task for 5-6 hours) * Determination of structure of organic compounds - NMR spectroscopy. (a task for 2 hours) * Task 2 will be realized at the Institute of Analytical Chemistry of the ASCR (see below for contact details).In the practice of advanced spectrometric methods, kinetic determination with UV / Vis detection technique will be demonstrated to determine trace amounts of copper.In another complex task, students will learn the methods of HPLC-ICP-MS and AAS, as well as sample pre-treatment.
First, mercury species will be extracted from both the sample and the reference material. Subsequently, the content of inorganically bound mercury and methylmercury in the extract will be determined by HPLC-ICP-MS.
The results will be compared with the determination of total mercury content without any pre-treatment of the sample on a dedicated atomic absorption spectrometer AMA-254.The last compulsory task of the practical exercise will be devoted to the use of NMR spectrometry for the identification of organic compounds based on 1H spectra.
This course is taught only in Czech.
