Syllabus

Molecular biology II:

The course represents series of advanced lectures on different methodologic aspects of modern molecular biology and builds upon the information presented and discussed in Molecular biology I. The typical student should have broad basic knowledge of biochemistry and physical chemistry. Intended for advanced students in the master course and for the PhD students of biochemistry and molecular biology. The course includes following topics: Recombinant DNA: isolation, preparation and manipulation with DNA (restriction enzymes, DNA polymerases, DNA-modifying enzymes. DNA libraries, detection and visualisation of DNA. Preparation of recombinant DNA). DNA sequencing (model organisms, genom procejts, sequencing of human genome: what we have learned?). PCR and its application.

Expression of recombinant proteins (bacteria, yeast, insect and mammalian cells. Detection and purification of the products). Protein engineering (random and site-directed mutagenesis. Structure-activity relationships, protein design de novo). Protein identification and interaction (monoclonal antibodies, protein-protein interaction in bacteria and yeast, two-hybrid system).

Combinatorics in modern biology (peptide and non-peptide libraries, gene chips, diferential expression arrays).

Mammalian cell as an experimental tool (immortalisation of cells, growth factors, stem cell research).

Transgenic animals (gene knock-outs, gene-modified organisms, gene technology in agriculture and medicine).

Social and ethical aspects of modern biotechnology and recombinant DNA: a blessing or a curse?

The course represents series of advanced lectures on different methodologic aspects of modern molecular biology and builds upon the information presented and discussed in Molecular biology I. The lectures cover principles of various recombinant DNA methods, DNA sequencing and genomics, expression and purification of recombinant proteins, transgenic animals and combinatorial approaches in modern biology. Ethical and social aspects of molecular biology are also discussed. The typical student should have broad basic knowledge of biochemistry and physical chemistry.
