Study on 99mTc-MAG3 and 99mTc-DMSA renal accumulation using in vitro cellular model

Abstract

99mTc-MAG3 and 99mTc-DMSA are renal radiodiagnostics in clinical use. However, the renal mechanisms determining their transport within the kidney are not fully explained.

The investigation compared renal uptake of the studied compounds and evaluated contribution of active and passive transport mechanisms to the renal accumulation. As the experimental model, freshly isolated rat renal cells were used.

The results documented significant quantitative and qualitative differences between the tested agents. The found data indicated that 99mTc-DMSA was transported across the membranes of the renal cells almost exclusively by passive transport.

On the other side, 99mTc-MAG3 accumulation rate in the rat renal cells was several times higher and was mediated by active transport mechanisms.
