Is rat liver affected by non-alcoholic steatosis more susceptible to the acute toxic effect of thioacetamide?

Abstract

There is only little evidence about altered sensitivity of steatotic liver to acute toxic injury. The aim of this project was to compare acute toxic effect of a model hepatotoxin thioacetamide (100 mg/ kg) on intact rat liver and rat liver affected by high-fat diet-induced non-alcoholic fatty liver disease (NAFLD).

Serum biochemical parameters (ALT, AST, ALP, and bilirubin) and TNFalpha, hepatic IL-6 and TGFbeta1 and centrilobular necrosis with subsequent regenerative response of the liver were significantly higher in high-fat diet fed rats after thioacetamide administration. Steatotic rat liver, compared to non-steatotic liver, is more sensitive to acute toxic effect of thioacetamide.
