Abstract

An optimized protocol for preparation of yeast cells for immunoelectron microscopy has been found, which preserves both their ultrastructure and antigenicity of sensitive nuclear antigens. This protocol is based on high-pressure freezing with a mixture of acetone and 0.1% uranyl actetate serving as cryo-substitution medium.
