Normal differentiation of hematopoietic cells is guided at the level of transcription during which specific transcription factors recognize specific DNA packaged in a spatial nucleoprotein structure referred to as chromatin. This process however is significantly deregulated during leukemogenesis resulting in a production of clonal highly proliferative cells blocked in differentiation.
Major regulators that play significant role in hematopoietic cell fate decision are transcription factors: GATA-1 and PU.1 and chromatin remodeling ATPase: Smarca5
