Remosomes: RSC generated non-mobilized particles with approximately 180 bp DNA loosely associated with the histone octamer

Abstract

Intermediate products generated during the process of chromatin remodeling by RSC complex, the so-called remosomes, were identified by high-resolution cryo-electron microscopy and molecular biology methods. This remosome (remodeled nucleosome) is a metastable product of the first phase of the remodeling process.

In contrast to original nucleosome, about 35 bp of DNA are associated with histone octamer, which results in creation of a DNA loop that dynamically moves around the perimeter of the octamer and allows thus access to originally inaccessible nucleosomal DNA. The measurement of the DNA accessibility by restricition enzymes showed that nucleosomal DNA was about 5x more accessible in comparison to its original state.
