New approaches to stimulation of bone formation

Abstract

Human genetics revealed the major role of Wnt signalling in bone formation and explained anabolic effect of intermittent parathyroid hormone on the skeleton and further development of analogs of PTH, PTHrP and calcilytics. Other approaches include monoclonal antibodies to powerful inhibitors of the Wnt signalling, namely sclerostin, the protein product of the SOST gene that is produced in osteocytes, and another inhibitory proteins.
