Abstract
New approach by two-photon excitation confocal microscopy to the whole yeast microcolony analysis allows the study of individual cells directly in situ. It revealed that 1) the S. cerevisiae colonies are covered with a thin protective skin-like surface cell layer and 2) Ato1p appears synchronously in cells, which do not originate from the same ancestor, but occupy specific position within the colony.
