The article provides a brief overview of BOLERO-2 study that evaluated the treatment of advanced HR+/HER2- breast cancer in postmenopausal women after previous treatment with nonsteroidal aromatase inhibitors (NSAIs) who were subsequently treated with the combination exemestane and everolimus. In particular, the subgroup of patients after previous treatment with anastrozole or letrozole in adjuvant setting or those in whom this treatment failed attained excellent median of progression-free survival (PFS) when treated with the combination exemestane + everolimus compared to exemestane in monotherapy.
Results from BOLERO trial are documented with the observation of an individual patient who was successfully treated with this combination. Everolimus is undoubtedly the first drug that has been shown to enhance the effect of hormonal therapy and delay resistance to hormonal therapy in postmenopausal patients with HR+/HER2- advanced breast cancer after previous NSAI failure.
