Role of percutaneous biopsy of skeletal muscle in diabetes research. Methodological summary

Abstract

Impaired skeletal muscle glucose disposal is a pathophysiological hallmark of type 2 diabetes. Easy and reproducible method of muscle sampling was needed for study of subcellular basis of its developement, i.e. percutaneous muscle biopsy.

This technique is nowaday a routine method of clinical diabetes research and has been described in details since seventhies of 20th century. Prof.

Vondra was one of the contributors of this method in our context and with his aid the method was established in our facility. The article deals with the technique of skeletal muscle sampling and possible analysis of the sample with regard to diabetes studies.

Methods of intramyocellular lipids assessement, genomics, proteomics, immunohistochemistry and mitochondrial biogenesis are described as well as satelite cells isolation and primary culture procedure.
