Rapid classification of unknown biological material using a novel triplex assay

Abstract

The methods employed for species-level identification of plant and animal material include DNA extraction, DNA quantification and Sanger sequencing of selected mitochondrial or plastid regions. In this study, samples without a clear origin (human/non-human/plant) are tested using a novel triplex PCR assay for rapid classification.

The assay enables detection of animal/human and plant DNA as well as the presence of PCR inhibitors. PCR products are separated using agarose electrophoresis.

In the case of positive detection of animal/human and/or plant material, the PCR product can be directly used for subsequent Sanger sequencing and species determination.
