Evaluation of retinal toxicity of carboplatin after repeated transcorneal intravitreal injection

Six albino rabbits were included in an in vivo experiment. Transcorneal intravitreal administrations of carboplatin in healthy albino rabbits were used as a model for possible clinical use in human intraocular retinoblastoma "seeding" tumor chemotherapy.

We administrated 0.05 mg dose of carboplatin intravitreally three times at two-week intervals, Electroretinograms (ERGs) were recorded before first administration of carboplatin and after last injection when also platinum concentration was measured. Reductions in the dark-adapted b-wave amplitudes and in the light-adapted b-wave and a-wave amplitudes and chorioretinal atrophy were noted after treatment.

We considered administered amount of carboplatin probably too high for treatment of retinoblastoma patients.
