HPLC-MS/MS analysis of degradation products of neosalvarsan in 76-year-old injection preparation

Abstract

High-performance liquid chromatography hyphenated with tandem mass spectrometry (LC-MS/MS) was used to analyze a 76-year-old injection preparation of neosalvarsan. The sample constituents were separated on the reverse phase C18 by the optimized gradient elution of methanol and water.

Using electrospray ionization in the negative mode (ESI-), the complete disintegration of neosalvarsan was proved. The structures of two degradation products of neosalvarsan, which are 2-(hydroxymethylamino)-4-arsenosophenol and As-(3-amino-4-hydroxyphenyl)arsonous acid were identified by MS2, and the ESI- fragmentation mechanism was proposed.
