Methodology of asymbiotic sowing of vanilla Vanilla planifolia in vitro

Abstract

Vanilla planifolia is probably the most important species of orchids in agriculture. However, only vegetative reproduction of this species is currently mastered.

For the purposes of agriculture, research and protection of wild populations of this species, there is a need for generative reproduction protocol, which can sustain genetic variability. This methodology describes currently the most effective protocol for germination of V. planifolia seeds in vitro.

It describes collection, storage, disinfection, sowing and cultivation conditions. This methodology introduces, for the first time, disinfection of orchid seeds with undiluted acid, specifically hydrochloric acid.

Furthermore, it describes relatively high temperature (30 °C) as a suitable temperature and medium BM1 as a good cultivation medium for germination of this Vanilla species. In comparison with previous reports, this methodology brings a great improvement to V. planifolia seed germination in in vitro conditions.
