Targeted treatment is characterized by high affinity and specificity against target structures, however, there are important specifics in pharmacokinetics and pharmacodynamics, which may impact clinical practice. Here we discuss difference between small and large molecules and in detail we focus on characteristics of monoclonal antibodies.
Kinetics of those large proteins in organism is different from small molecular drugs. We also discuss some of the factors which may influence the therapeutic effect of antibodies, e.g. body weight (obesity), anti-drug antibody formation, drug interactions or some pathological states and examples focused mainly on anti-IgE, anti-IL5 or anti-IL5R antibodies.
